After the three successive clusterings and the identification of the kinetic families discriminating between the prime and boost responses, the last step of our longitudinal mass cytometry data analysis was to compare the marker expression between the two classes. We identified and ranked the markers differentially expressed between ‘primary‘ and ‘secondary‘ granulocytes and monocytes-DC, based on the Kolmogorov-Smirnov distance.
As cytometrists and immunologists, we ultimately want to see histograms! The expression patterns of top markers can be displayed in the different phenotypic families.
