As the first analysis step, macaque leukocytes with similar phenotypes were grouped together into cell clusters using the Spanning-tree Progression Analysis of Density-normalized Events (SPADE) algorithm.

This tree includes 900 clusters, each containing cells sharing similar phenotypes. Phenotypically similar cell clusters are connected in a minimum spanning tree.

Note that all the cells from all the samples (all animals and all timepoints) were used to build the tree.

The SPADE tree can be colored based on the expression of 2 markers used for NK cell isolation. An annotated SPADE tree is also provided.

NK cell SPADE clusters were isolated as CD3- CD8+ cells.
